Overview

Dostarlimab + niraparib is a combination therapy consisting of two distinct agents used primarily in oncology. Dostarlimab is a monoclonal antibody that acts as an immune checkpoint inhibitor by targeting the programmed death 1 (PD-1) receptor, thereby enhancing the immune system's ability to recognize and attack tumor cells. Niraparib is a small molecule inhibitor of poly (ADP-ribose) polymerase (PARP), an enzyme involved in DNA repair; its inhibition leads to accumulation of DNA damage and cancer cell death, particularly in tumors with defective homologous recombination repair mechanisms such as BRCA mutations. The combination has been investigated for synergistic antitumor effects, especially in recurrent or advanced cancers including ovarian, endometrial, breast, and penile squamous cell carcinoma[1][3][4][6]. Clinical trials have shown improved progression-free survival with this regimen compared to standard therapies[3].
