Overview

DOTA-biotin-avidin (DBA) is an experimental radiotherapeutic conjugate designed for the intravesical treatment of non-muscle invasive bladder cancer (NMIBC). The system utilizes a pretargeting-inspired approach that leverages the exceptionally high affinity between avidin and biotin to form a large molecular complex. When radiolabeled with a beta-emitting isotope such as yttrium-90 (90Y) via the DOTA chelator, the resulting [90Y]DBA complex is administered directly into the bladder. The size and biochemical properties of the avidin-biotin-DOTA conjugate prevent its re-absorption through the bladder wall into the systemic circulation, thereby localizing the short-range radiation to the tumor tissue and urothelium while minimizing systemic toxicity. Preclinical studies in murine models have demonstrated that a single dose of [90Y]DBA significantly reduces tumor growth without causing irreversible damage to the normal bladder morphology.
