Overview

DOTA-C595 is a radioimmunoconjugate consisting of the monoclonal antibody C595, which targets the cancer-specific mucin 1 epitope (MUC1-CE), conjugated to the chelator DOTA (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid). It is designed to be labeled with radionuclides such as Lutetium-177 ([177Lu]Lu-DOTA-C595) or Actinium-225 ([225Ac]Ac-DOTA-C595) for the treatment of malignancies overexpressing MUC1, particularly pancreatic ductal adenocarcinoma (PDAC). The antibody component provides high-affinity binding to MUC1-CE, while the radionuclide delivers targeted radiation (beta or alpha particles) to induce DNA damage and cell death in tumor cells. Research has demonstrated that the alpha-emitting version, [225Ac]Ac-DOTA-C595, can overcome typical treatment barriers like hypoxia due to the high linear energy transfer of alpha particles.
