Overview

Double-loaded dendritic cell therapy (DOC1021) is a personalized autologous cancer immunotherapy developed by Diakonos Oncology. The platform utilizes a unique 'double-loading' technique where patient-derived dendritic cells are simultaneously pulsed with both tumor lysate and tumor-derived mRNA. This dual-loading strategy is designed to mimic a viral infection, programming both MHC class I and MHC class II pathways to trigger a robust and comprehensive immune response involving both CD8+ cytotoxic T cells and CD4+ helper T cells. By targeting a broad spectrum of patient-specific tumor-associated antigens (uTAAs), the therapy aims to overcome the limitations of single-antigen vaccines. It is currently being evaluated in clinical trials for glioblastoma, ovarian cancer, and acute myeloid leukemia (AML), and is administered via injection near lymph nodes to facilitate rapid migration to the immune system.
