double recombinant vaccinia virus VV-GMCSF-Lact is a genetically engineered oncolytic virus based on the Lister strain (L-IVP) of the vaccinia virus. It has deletions in the viral thymidine kinase and growth factor genes, with insertions of two therapeutic genes: human granulocyte-macrophage colony-stimulating factor (GM-CSF) and lactaptin, an apoptosis-inducing protein derived from human milk kappa-casein. GM-CSF stimulates antitumor immune responses by recruiting granulocytes, macrophages, and promoting dendritic cell differentiation. Lactaptin induces apoptosis in cancer cells via the mitochondrial pathway. The drug is designed for intratumoral or intravenous administration to treat solid tumors such as glioma and metastatic breast cancer, showing high cytotoxic activity against chemoresistant tumor models[2][5][6].
