Double-stranded RNA-clNPs (dsRNA-clNPs) are an experimental immunotherapeutic formulation consisting of long double-stranded RNA (dsRNA) complexed with cationic polystyrene latex nanostructures (clNPs). The clNPs serve as a delivery vehicle to enhance the cellular uptake and stability of the dsRNA, which is otherwise susceptible to degradation. Once internalized by target cells, such as ovarian cancer cells, the dsRNA acts as a potent agonist for innate immune sensors including Toll-like receptor 3 (TLR3), RIG-I, and MDA5. This activation triggers a robust type I interferon response and the expression of interferon-stimulated genes (ISGs), effectively inducing an antiviral-like state that can potentiate the body's immune response against tumors. Research indicates that the clNP delivery system significantly improves the immunostimulatory capacity of dsRNA compared to uncomplexed nucleic acids.
