Overview

DOV 216,303 is an experimental antidepressant classified as a triple reuptake inhibitor (TRI), also known as a serotonin-norepinephrine-dopamine reuptake inhibitor (SNDRI). It acts by inhibiting the reuptake of serotonin (SERT), norepinephrine (NET), and dopamine (DAT) with reported IC50 values of approximately 14 nM for SERT, 20 nM for NET, and 78 nM for DAT. The compound is a racemic mixture composed of amitifadine (DOV-21,947) and its (-)-enantiomer DOV-102,677. Originally developed by DOV Pharmaceutical and later licensed to Merck & Co., development was discontinued after patent expiration in the late 2000s. Clinical studies demonstrated that it was safe and well-tolerated at doses up to at least 100 mg/day over short periods in humans but caused some gastrointestinal disturbances at higher doses. Preclinical research indicated potential utility not only in depression but also in recovery from spinal cord injury and possibly obesity due to effects on plasma triglycerides[1][5][7][10].
