Dovitinib + abiraterone acetate + prednisone is a combination therapy regimen evaluated for the treatment of metastatic castration-resistant prostate cancer (mCRPC). Dovitinib (TKI258) is a multi-targeted small molecule tyrosine kinase inhibitor that primarily targets fibroblast growth factor receptors (FGFR1, FGFR2, and FGFR3), vascular endothelial growth factor receptors (VEGFR1, VEGFR2, and VEGFR3), and platelet-derived growth factor receptor beta (PDGFRB). Abiraterone acetate is an androgen biosynthesis inhibitor that selectively and irreversibly inhibits the enzyme CYP17A1 (17α-hydroxylase/17,20-lyase), thereby blocking the production of testosterone in the testes, adrenal glands, and prostate tumor tissue. Prednisone, a corticosteroid, is co-administered with abiraterone to compensate for the reduction in serum cortisol and to mitigate mineralocorticoid excess. This combination aims to provide a dual-pronged attack by inhibiting both the androgen receptor signaling pathway and growth factor-mediated pathways that often contribute to treatment resistance in advanced prostate cancer.
