Drug intelligence / Profile preview
Downtuss AD is a combination oral liquid formulation developed by Healthkind Labs for the symptomatic relief of cough, cold, and upper respiratory tract congestion. It contains five active ingredients: dextromethorphan (a cough suppressant), ambroxol (a mucolytic agent), chlorpheniramine (an antihistamine), phenylephrine (a nasal decongestant), and menthol (a cooling agent). Dextromethorphan acts as a sigma-1 receptor agonist and NMDA receptor antagonist to suppress the cough reflex. Ambroxol reduces mucus viscosity and enhances mucociliary clearance, while also blocking voltage-gated sodium channels (Nav1.7 and Nav1.8) to provide local anesthetic effects. Chlorpheniramine competitively inhibits histamine H1 receptors to alleviate allergic symptoms such as sneezing and rhinorrhea. Phenylephrine is a selective alpha-1 adrenergic receptor agonist that induces vasoconstriction in the nasal mucosa, reducing swelling and congestion. Menthol acts as a TRPM8 agonist, providing a cooling sensation that helps soothe throat irritation.
