Overview

Doxepin is a tricyclic antidepressant (TCA) used primarily to treat major depressive disorder, anxiety disorders, and insomnia. It is also sometimes used as an alternative treatment for chronic hives when other antihistamines are ineffective. Doxepin works mainly by inhibiting the reuptake of norepinephrine and serotonin in the brain, thereby increasing their levels and improving mood balance[1][2][4][8]. At lower doses (such as those used for insomnia), it acts predominantly as a highly selective antagonist of the histamine H1 receptor, which promotes sleep maintenance[6]. Doxepin also exhibits anticholinergic, antiadrenergic, antiserotonergic effects and can block voltage-gated sodium channels[7]. The drug was originally developed by Pfizer.
