Doxifluridine + medroxyprogesterone acetate is a combination therapy consisting of two active pharmaceutical ingredients used primarily in the treatment of advanced or recurrent breast cancer, particularly in cases resistant to anthracycline-based regimens. Doxifluridine is an oral prodrug of 5-fluorouracil (5-FU), a pyrimidine analog that inhibits DNA synthesis by interfering with thymidylate synthase, leading to cytotoxic effects on rapidly dividing tumor cells. Medroxyprogesterone acetate is a synthetic progestin hormone with antitumor activity, thought to exert its effect through modulation of hormone receptors and inhibition of tumor cell proliferation. This combination has been studied as part of chemoendocrine regimens for patients with advanced breast cancer, often alongside other agents such as mitoxantrone or cyclophosphamide[2][3][4]. The regimen aims to leverage both cytotoxic and hormonal mechanisms for improved efficacy.
