Bleomycin is a glycopeptide antibiotic that binds DNA and induces single- and double-strand breaks via free radical formation, causing cell cycle arrest primarily in G2. Vincristine is a vinca alkaloid that binds β-tubulin and inhibits microtubule polymerization, arresting cells in metaphase. The combination is used in oncology settings where overlapping activity against rapidly dividing cells is desired; historically related regimens appear in lymphomas and solid tumors, though modern standards often prefer other combinations for specific indications.
