This combination chemotherapy regimen, consisting of doxorubicin, cyclophosphamide, and cisplatin, is being investigated by West China Hospital in a Phase 2 clinical trial for the treatment of locally advanced or metastatic poorly differentiated or anaplastic thyroid cancer. Doxorubicin is an anthracycline that intercalates into DNA and inhibits topoisomerase II; cyclophosphamide is an alkylating agent that forms DNA cross-links; and cisplatin is a platinum-based antineoplastic agent that creates DNA adducts. Together, these agents aim to inhibit DNA replication and induce apoptosis in aggressive thyroid malignancies.
