Overview

This is a four-drug combination chemotherapy regimen consisting of doxorubicin (an anthracycline antibiotic), cyclophosphamide (an alkylating agent), methotrexate (an antimetabolite and antifolate agent), and procarbazine (a methylhydrazine derivative alkylating agent). The combination, often referred to as the CAMP regimen, has been used primarily in the treatment of metastatic non-oat cell bronchogenic carcinoma (non-small cell lung cancer) and has also been explored for use in lymphomas. Each drug works via a distinct mechanism to inhibit cancer cell proliferation or induce apoptosis. Doxorubicin intercalates DNA and inhibits topoisomerase II; cyclophosphamide crosslinks DNA; methotrexate inhibits dihydrofolate reductase, blocking DNA synthesis; and procarbazine causes DNA strand breaks through alkylation. The rationale for combining these agents is to maximize anti-tumor efficacy by targeting different pathways involved in tumor growth while minimizing resistance[1][2][3].
