Doxorubicin + fluorouracil + methotrexate is a combination chemotherapy regimen, commonly referred to as FAMTX. It consists of three cytotoxic agents: - Doxorubicin, an anthracycline antibiotic that intercalates DNA and inhibits topoisomerase II, leading to DNA strand breaks and apoptosis. - Fluorouracil (5-FU), a pyrimidine analog that inhibits thymidylate synthase, disrupting DNA synthesis and repair. - Methotrexate, an antimetabolite that inhibits dihydrofolate reductase (DHFR), blocking folic acid metabolism required for nucleotide synthesis. This combination has been used primarily in the treatment of advanced gastric cancer and has also been studied in other solid tumors such as breast cancer. The regimen aims to exploit the synergistic cytotoxic effects of these agents on rapidly dividing tumor cells[2][3][4].
