doxorubicin + Genexol-PM is a combination chemotherapy regimen consisting of the anthracycline doxorubicin and Genexol-PM, a Cremophor EL-free, polymeric micellar formulation of paclitaxel. Developed by Samyang Biopharmaceuticals, Genexol-PM utilizes a biodegradable amphiphilic diblock copolymer (mPEG-PDLLA) drug delivery vehicle to encapsulate paclitaxel, forming stable micelles that improve solubility and reduce the hypersensitivity risks associated with traditional paclitaxel formulations. Doxorubicin acts by intercalating DNA and inhibiting topoisomerase II, which leads to DNA strand breaks and cell death. Paclitaxel, the active ingredient in Genexol-PM, stabilizes microtubules to inhibit their disassembly, thereby arresting the cell cycle in the G2/M phase. This combination has been evaluated in Phase II clinical trials by Gachon University Gil Medical Center for the treatment of advanced or metastatic HER2-negative breast cancer, demonstrating significant objective response rates.
