This is a combination chemotherapy regimen consisting of three cytotoxic agents—doxorubicin, mitomycin (specifically as mitomycin C), and vinblastine. Each drug has a distinct mechanism of action: - Doxorubicin is an anthracycline antibiotic that intercalates DNA and inhibits topoisomerase II, leading to DNA strand breaks and inhibition of replication. - Mitomycin C is an alkylating agent that crosslinks DNA, inhibiting DNA synthesis and function. - Vinblastine is a vinca alkaloid that binds to tubulin, inhibiting microtubule formation and thereby arresting cell division at metaphase. The combination has been studied primarily in the treatment of refractory or metastatic breast cancer. Clinical studies have shown response rates around 33% in heavily pretreated patients with metastatic breast carcinoma. Toxicities are mainly hematologic (notably bone marrow suppression), but the regimen was generally tolerable in clinical trials[1][3]. The contribution of each component to overall efficacy remains unclear due to overlapping mechanisms with other regimens[1].
