Overview

This is a combination regimen of three drugs—doxorubicin, paclitaxel, and trastuzumab—used primarily in the treatment of HER2-positive breast cancer. Doxorubicin is an anthracycline chemotherapy that intercalates DNA and inhibits topoisomerase II, leading to DNA damage and cell death. Paclitaxel is a taxane chemotherapy that stabilizes microtubules, preventing cell division. Trastuzumab is a monoclonal antibody targeting the human epidermal growth factor receptor 2 (HER2), blocking its signaling and marking HER2-overexpressing cancer cells for immune-mediated destruction. This combination exploits both cytotoxic effects on rapidly dividing tumor cells (doxorubicin and paclitaxel) and targeted inhibition of HER2-driven proliferation (trastuzumab). It is used as adjuvant or neoadjuvant therapy for early-stage or locally advanced HER2-positive breast cancers[1][4][6][7].
