A combination of two chemotherapeutic agents - doxorubicin, an anthracycline antibiotic, and temozolomide, an alkylating agent capable of crossing the blood-brain barrier. This combination has demonstrated synergistic effects in inhibiting cancer cells, particularly in glioblastoma multiforme (GBM), including temozolomide-resistant cells. The combination enhances doxorubicin uptake and induces higher apoptosis rates. Researchers have also developed conjugated compounds coupling these drugs via an acylhydrazone linkage as potential prodrugs for GBM treatment.
