Overview

This is a combination chemotherapy regimen used in the treatment of advanced adrenocortical carcinoma (ACC). The regimen consists of doxorubicin (an anthracycline antibiotic), vincristine (a vinca alkaloid), etoposide (a topoisomerase inhibitor), and mitotane (an adrenocortical cytotoxic agent). Mitotane is often administered orally at doses up to 4 g/day with the goal of attaining blood levels of 14-20 mg/L, while the other components are administered intravenously. This combination has been studied in clinical trials for patients with metastatic adrenocortical carcinoma not amenable to radical surgery.
