Drug intelligence / Profile preview
Doxorubicin glycoconjugate refers to novel oxime-linked doxorubicin-glucose conjugates (specifically EB73 and EB74) developed by Meyer Children's Hospital IRCCS and the University of Florence. These conjugates are designed to improve the delivery of the chemotherapeutic agent doxorubicin across the blood-brain barrier (BBB) to target glioblastoma (GBM). By conjugating doxorubicin to glucose, the drug exploits the Warburg effect and the overexpression of glucose transporter 1 (GLUT1) on both the BBB and malignant brain tumor cells to enhance cellular uptake and selectivity. Preclinical studies have demonstrated that these glycoconjugates maintain the DNA intercalation capability of doxorubicin while showing enhanced cytotoxicity and pro-apoptotic effects in glioblastoma cell lines, including doxorubicin-resistant cells.
