Drug intelligence / Profile preview
Doxorubicin HCl liposome (brand name: Doxil) is a pegylated liposomal formulation of doxorubicin hydrochloride, a cytotoxic anthracycline antibiotic. The active ingredient is encapsulated in microscopic phospholipid bilayer vesicles coated with methoxypolyethylene glycol (MPEG), known as STEALTH liposomes. This pegylation protects the drug from immune system detection and significantly extends its blood circulation time, allowing for preferential accumulation in tumor tissues via the enhanced permeability and retention (EPR) effect. Its mechanism of action involves intercalating into DNA and inhibiting topoisomerase II, which disrupts DNA replication and causes cell death. It is FDA-approved for the treatment of ovarian cancer, AIDS-related Kaposi's sarcoma, and multiple myeloma, and is investigated for other solid tumors including breast and endometrial cancers.
