12b80 is an experimental antineoplastic agent designed for targeted treatment of bone cancers, particularly osteosarcoma. It is a prodrug consisting of the chemotherapeutic agent doxorubicin covalently bound to a bone-targeting hydroxybisphosphonate vector via a pH-sensitive linker. This design enables selective release of doxorubicin in the acidic microenvironment typical of bone tumors, thereby increasing drug delivery to tumor sites while reducing systemic toxicity. Preclinical studies have demonstrated that 12b80 achieves higher concentrations in bone tissue and tumor-associated heterotopic bone compared to non-targeted doxorubicin, with improved safety and efficacy profiles. The drug was developed by Atlanthera Drug Discovery Company (France) and has shown promising results in animal models as well as early-phase clinical trials for osteosarcoma[1][2][3][4].
