Drug intelligence / Profile preview
LEAF-1404 is a complex generic version of pegylated liposomal doxorubicin (originally marketed as Caelyx/Doxil). Developed by LEAF Pharmaceuticals and manufactured by its subsidiary LEAF Rwanda, LEAF-1404 is designed to treat ovarian cancer, breast cancer, and AIDS-related Kaposi's sarcoma. The formulation consists of doxorubicin hydrochloride encapsulated within polyethylene glycol (PEG)-coated liposomes, which alters the drug's pharmacokinetics by prolonging circulation time and reducing cardiotoxicity compared to conventional doxorubicin. Once released, doxorubicin acts as an anthracycline topoisomerase II inhibitor and DNA intercalator, disrupting DNA replication and transcription to induce cancer cell death.
