Overview

Doxycycline + atorvastatin is an investigational dual-drug nanoparticle formulation developed by Fayoum University for the local treatment of chronic periodontitis. The formulation consists of doxycycline, a tetracycline antibiotic and matrix metalloproteinase (MMP) inhibitor, and atorvastatin, a statin with anti-inflammatory and bone-regenerative properties, co-encapsulated in chitosan nanoparticles. This drug delivery system is designed for sustained release within periodontal pockets as an adjunct to scaling and root planing (SRP). Doxycycline provides antimicrobial activity against periodontal pathogens and inhibits MMP-8 to prevent connective tissue destruction, while atorvastatin promotes osteogenesis and reduces inflammation. It is currently being evaluated in a Phase 4 clinical trial for adults with stage II and III chronic periodontitis.
