Doxycycline + citric acid + polysorbate 80 (often referred to as the DCP lock solution or GLP-2500) is an investigational antimicrobial lock solution (ALS) specifically designed for the prevention and treatment of catheter-related bloodstream infections (CRBSI). The formulation consists of three functional components: doxycycline, a tetracycline antibiotic that provides broad-spectrum antibacterial activity by inhibiting protein synthesis via the 30S ribosomal subunit; citric acid, which acts as a calcium-chelating anticoagulant to maintain catheter patency and as an acidifier to enhance antibiotic stability and efficacy; and polysorbate 80, a non-ionic surfactant that disrupts and penetrates microbial biofilms. By combining these agents, the solution is able to eradicate sessile bacteria and fungi protected within the biofilm matrix of indwelling catheters, which are otherwise resistant to systemic antibiotic therapy. It has been primarily investigated for use in high-risk patients, such as those undergoing long-term hemodialysis or oncological treatment.
