Overview

Doxycycline + rifampicin + ceftriaxone is a combination antimicrobial regimen utilized for the treatment of complicated brucellosis, particularly moderate-to-severe osteoarticular brucellosis. The regimen consists of two oral agents, **doxycycline** (a tetracycline) and **rifampicin** (a rifamycin), combined with the parenteral administration of **ceftriaxone** (a third-generation cephalosporin). Doxycycline inhibits bacterial protein synthesis by binding to the 30S ribosomal subunit, rifampicin inhibits bacterial RNA polymerase, and ceftriaxone inhibits penicillin-binding proteins involved in cell wall synthesis. This triple therapy is designed to provide synergistic antibacterial activity and enhanced tissue penetration to target joint and bone lesions, aiming to improve cure rates and reduce the risk of recurrence in severe cases of *Brucella* infection.
