DP-001 (also known as 2MD or 2-methylene-19-nor-(20S)-1α,25-dihydroxyvitamin D3) is an oral small molecule vitamin D analog and potent 1α-hydroxylated vitamin D receptor (VDR) agonist. Developed by Deltanoid Pharmaceuticals in collaboration with the University of Wisconsin-Madison, it was primarily investigated for the treatment of secondary hyperparathyroidism in patients with chronic kidney disease on hemodialysis. The drug works by binding to the VDR and suppressing the synthesis and secretion of parathyroid hormone (PTH). Clinical trials demonstrated its efficacy in significantly reducing PTH levels with a longer half-life (approximately 50-56 hours) compared to existing vitamin D therapies. However, development appears to have ceased following the closure of Deltanoid Pharmaceuticals in 2018.
