Overview

DP-9149 is an investigational small molecule activator of GCN2 (General Control Nonderepressible 2) developed by Deciphera Pharmaceuticals. It functions by directly binding to and paradoxically activating the GCN2 kinase, a key component of the Integrated Stress Response (ISR) pathway. This activation leads to the upregulation of downstream ISR components, including ATF4 and CHOP, which triggers programmed cell death in oncogene-driven solid tumor cells. Preclinical data indicate significant activity in RAS-mutant and VHL-mutant cancers, both as a monotherapy and in combination with standard-of-care agents.
