Drug intelligence / Profile preview
DP-9244 is a potent and selective small molecule inhibitor of vacuolar protein sorting 34 (VPS34), a class III phosphoinositide 3-kinase (PI3K) that regulates autophagy and endosomal trafficking. Originally developed by Sprint Bioscience and licensed to Deciphera Pharmaceuticals, DP-9244 is designed to prevent the degradation of activated STING (stimulator of interferon genes) by inhibiting VPS34-mediated endosomal trafficking. In preclinical models, combining DP-9244 with a STING agonist (such as ADU-S100) delays STING degradation, prolongs downstream signaling (including pTBK1), and enhances type I interferon-mediated gene expression and cytokine secretion (such as CCL5 and CXCL10). This combination has demonstrated enhanced anti-tumor efficacy and tumor growth control in vivo, providing a therapeutic rationale for combining VPS34 inhibitors with STING agonists to treat solid tumors.
