DP47-muIL2v is an untargeted control immunocytokine developed by Roche (Genentech) for use in preclinical research, specifically in murine syngeneic tumor models. It consists of a murine interleukin-2 variant (muIL2v) fused to a non-targeting antibody scaffold, DP47. The IL-2 variant is engineered to lack binding to the IL-2Rα (CD25) subunit, which reduces the stimulation of regulatory T cells and associated toxicity, while maintaining affinity for the IL-2Rβγ (CD122/CD132) receptor complex on effector T cells and natural killer (NK) cells. Because the DP47 scaffold does not bind to any specific tumor antigen, DP47-muIL2v serves as a negative control to demonstrate the necessity of tumor-antigen targeting for the efficacy of targeted immunocytokines like muCEA-muIL2v or muFAP-muIL2v. It is primarily used to establish the baseline biological activity of the IL-2v moiety in the absence of tumor localization.
