DP7 is an antimicrobial and immunomodulatory peptide with the sequence VQWRIRVAVIRK. It is being investigated for its therapeutic potential in various conditions. In inflammatory bowel diseases, DP7 has been shown to ameliorate inflammation, attenuate intestinal barrier dysfunction, and modulate the gut microbiota. Derivatives of DP7, such as the cholesterol-modified cationic cell-penetrating peptide DP7-C, are utilized for enhanced delivery and specific therapeutic applications. DP7-C has been explored for promoting bone regeneration, partly by mediating macrophage-derived exosomal miR-21b, and as a systemic delivery system for siDR6 to treat metastatic lung cancer, where it inhibits tumor cell growth by suppressing the STAT3 signaling pathway and preventing metastasis. Another derivative, KK2DP7, a dendrimer-structured peptide, has demonstrated efficacy in anti-tumor therapy by stimulating CD11b+ cell populations in the spleen to elicit trained immunity through TLR2-IRF7 axis activation and epigenetic reprogramming.
