Overview

DPA-713 is a second-generation radioligand designed to target the translocator protein (TSPO), an 18 kDa protein situated on the outer mitochondrial membrane. TSPO is a well-established biomarker for inflammation, as its expression is significantly upregulated in activated microglia in the central nervous system and activated macrophages in peripheral tissues. DPA-713 is primarily utilized as a positron emission tomography (PET) imaging agent, most commonly labeled with Carbon-11 ([11C]DPA-713), to visualize and quantify inflammatory processes. It is used in clinical research to assess disease activity and treatment response in conditions such as rheumatoid arthritis, multiple sclerosis, and various neurodegenerative disorders. Compared to first-generation TSPO tracers, DPA-713 exhibits improved binding affinity and a superior signal-to-noise ratio, although its binding is sensitive to a common genetic polymorphism (rs6971) in the TSPO gene.
