dPD-1 (TSHR+CD19) CAR-T cells are an autologous chimeric antigen receptor (CAR) T-cell therapy designed to target thyroid-stimulating hormone receptor (TSHR) and CD19. Developed by the Third Affiliated Hospital of Sun Yat-sen University, this cell therapy is specifically investigated for the treatment of refractory or relapsed thyroid cancer. The construct incorporates a dominant-negative PD-1 (dPD-1) modification, which is intended to shield the CAR-T cells from PD-1/PD-L1 mediated immunosuppression within the tumor microenvironment, thereby enhancing their anti-tumor activity and persistence. The dual-targeting approach and the checkpoint blockade integration represent a strategy to improve efficacy in solid tumors like thyroid carcinoma.
