dPD-1 hCD19 CAR-T is a chimeric antigen receptor T cell (CAR-T) therapy designed to treat relapsed or refractory non-Hodgkin lymphoma and other B-cell malignancies. This "armored" CAR-T product is engineered to express a humanized anti-CD19 chimeric antigen receptor along with a dominant-negative PD-1 (dPD-1) component. The CD19 targeting enables the modified T cells to recognize and kill malignant B cells expressing CD19. The inclusion of the dominant-negative PD-1 element is intended to block the inhibitory effects of the PD-1/PD-L1 pathway on T cells within the tumor microenvironment, thereby enhancing antitumor activity and persistence of the infused cells. Developed by Innovative Cellular Therapeutics, this therapy represents an advanced approach in immunotherapy by combining direct tumor targeting with checkpoint inhibition in a single cellular product[1].
