Overview

DPI-125 is a novel, small-molecule, diarylmethylpiperazine compound that acts as a **triple agonist at opioid receptors** (delta, mu, and kappa). It demonstrates balanced agonism with the highest potency at the delta-opioid receptor, followed by similar potency for the mu- and kappa-opioid receptors. DPI-125 is being primarily developed for the treatment of **opioid use disorder** and has also been studied as an analgesic. Preclinical data indicate that DPI-125 has strong analgesic activity, a higher respiratory safety profile compared to traditional μ-agonist opioids, and lower abuse liability due to its balanced activity at all three opioid receptors. Unlike conventional opioids, DPI-125's unique receptor profile offers analgesia with reduced risk of respiratory depression and reduced reinforcing (addictive) effects. The drug is being developed by DMK Pharmaceuticals, following initial discovery by Enhance Biotech[1][2][3][4][5].
