Overview

DPTX3496 is an oral, small molecule condensate modulator (c-mod) developed by Dewpoint Therapeutics for the treatment of Wnt-driven cancers, including colorectal cancer, triple-negative breast cancer (TNBC), and non-small cell lung cancer (NSCLC)[1][3][4][6]. The drug targets beta catenin by sequestering it into nuclear condensate depots, thereby inhibiting its aberrant transcriptional activity and down-regulating Wnt pathway signaling. This mechanism selectively induces apoptosis in cancer cells with excessive beta catenin/Wnt signaling[3][4][6]. Preclinical studies have shown robust tumor regression and stasis in various Wnt-driven tumor models with good tolerability[3][4]. DPTX3496 is currently in IND-enabling studies as of early 2025, with a planned IND filing for the second half of 2025[3][4].
