DPX-BRAF + cyclophosphamide is an investigational cancer immunotherapy regimen consisting of DPX-BRAF, a depot-formulated therapeutic vaccine targeting BRAF-derived peptides, administered together with low-dose cyclophosphamide as an immune-modulating chemotherapeutic. DPX-BRAF is based on the DPX (DepoVax) platform, which presents tumor-associated antigens in a lipid-in-oil formulation designed to induce durable, antigen-specific CD4+ and CD8+ T-cell responses against BRAF-mutant tumor cells. Low-dose cyclophosphamide is used primarily for its immunomodulatory effect, particularly depletion or functional inhibition of regulatory T cells, thereby enhancing vaccine-induced anti-tumor T-cell activity. The combination is being developed for BRAF-mutant solid tumors, leveraging vaccine-induced T-cell cytotoxicity together with cyclophosphamide-mediated modulation of the tumor immune microenvironment.[7]
