**DPX-Survivac + pembrolizumab + cyclophosphamide** is an investigational multi-agent immunotherapy regimen combining DPX-Survivac (also known as maveropepimut-S), pembrolizumab (Keytruda), and cyclophosphamide. DPX-Survivac is a peptide-based cancer vaccine developed using the DepoVax (DPX) lipid nanoparticle platform, targeting survivin, a tumor-associated antigen highly expressed in multiple cancers. The vaccine is designed to induce robust and sustained survivin-specific T cell responses, resulting in immune-mediated tumor cell death. Pembrolizumab is a programmed death-1 (PD-1) checkpoint inhibitor monoclonal antibody, which blocks the interaction between PD-1 and its ligands to reverse T cell exhaustion and support anti-tumor immunity. Cyclophosphamide, given in low/intermittent dosing in this regimen, acts as an immunomodulator, mainly through regulatory T-cell (Treg) depletion and enhancement of vaccine-induced T-cell responses. The combination aims to leverage vaccine-induced, tumor-specific T-cells, checkpoint blockade to overcome immune evasion, and cyclophosphamide's immunomodulatory effects, primarily for advanced ovarian, fallopian tube, and peritoneal cancers.
