DR-01 is a non-fucosylated human immunoglobulin G1 (IgG1) monoclonal antibody that targets CD94, a receptor selectively expressed on cytotoxic terminal effector CD8+ T cells, γδT cells, and natural killer (NK) cells. By binding to CD94, DR-01 leads to rapid depletion of these target cells primarily through antibody-dependent cellular cytotoxicity (ADCC), including fratricide. The drug is being developed by Dren Bio for the treatment of large granular lymphocytic leukemia (LGLL) and various cytotoxic lymphomas—including aggressive NK cell leukemia, cutaneous T-cell lymphoma (CTCL), EBV T-cell lymphoma, hepatosplenic T-cell lymphoma—as well as other related hematologic malignancies. It is currently in Phase 1/2 clinical trials for relapsed or refractory cases of these diseases.
