DR30303 is a humanized anti-Claudin 18.2 (CLDN18.2) heavy chain antibody Fc fusion protein developed using Doer Bio's SMART-VHHBody platform[1][2][3]. It is composed of a single domain (VHH) antibody fused to an engineered human IgG1 Fc region[3][5]. The drug specifically targets and binds to CLDN18.2, a tight junction protein abnormally expressed on certain tumor cells, including those in gastric and other gastrointestinal cancers[1][3]. Upon binding to CLDN18.2-expressing tumor cells, DR30303 induces antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC), leading to the destruction of these cancer cells while minimizing toxicity to normal tissues expressing CLDN18.2[1][3]. The primary indications under investigation are advanced or metastatic solid tumors with abnormal expression of CLDN18.2, including gastric cancer, esophageal cancer, pancreatic cancer, and other digestive system neoplasms[3][4][5].
