Overview

DR510 is an epidermal growth factor receptor (EGFR)-targeting T-cell engager prodrug developed by Zhejiang Doer Biologics Co., Ltd. It is designed to overcome critical challenges in solid tumor treatment, such as on-target off-tumor toxicities, cytokine release syndrome (CRS), and immune effector cell-associated neurotoxicity syndrome (ICANS). The drug leverages a novel cleavable substrate linker and a SP34-derived anti-CD3 antibody VHH masking domain (VHHm). An albumin-binding domain (ABD) is fused to VHHm, which 'locks' the moderate-affinity anti-EGFR Fab through interaction with the anti-CD3 scFv, achieving dual masking of both functional domains. This unique single-site cleavage mechanism for ABD-VHHm release in circulation distinguishes it from previously reported TCE prodrugs. DR510 aims to achieve a critical balance between therapeutic potency and safety for the treatment of EGFR-positive solid tumors.
