Overview

This entry refers to a Phase 3 clinical trial (NCT01942343) conducted by the University Hospital of Strasbourg, France, evaluating the incidence of akathisia when using droperidol versus ondansetron for the prevention of postoperative nausea and vomiting (PONV) in ambulatory surgery. Droperidol is a butyrophenone-derived dopamine D2 receptor antagonist, while ondansetron is a selective 5-HT3 receptor antagonist. The study compared two low doses of droperidol (0.625 mg and 1.25 mg) against a standard 4 mg dose of ondansetron to determine if droperidol increased the risk of akathisia, a movement disorder characterized by inner restlessness, in the postoperative period. The trial found that both medications were associated with a low and comparable incidence of akathisia, supporting the safety profile of low-dose droperidol in the outpatient setting.
