Overview

DS-1055a is a novel afucosylated human monoclonal antibody that targets GARP (Glycoprotein-A Repetitions Predominant), a transmembrane protein expressed on the surface of activated regulatory T cells (Tregs) in the tumor microenvironment. By binding to GARP, DS-1055a selectively depletes GARP-positive Tregs, reversing immunosuppression and enhancing anti-tumor immune responses. This mechanism aims to restore antitumor immunity in patients with various cancers, including those resistant or refractory to checkpoint inhibitors. The drug is being developed primarily for use in relapsed or refractory advanced or metastatic solid tumors such as head and neck cancer, gastric cancer, esophageal cancer, melanoma, non-small-cell lung cancer, and other solid tumors[1][2][4][5][6].
