Overview

DS-1103a is a recombinant humanized IgG4 monoclonal antibody that targets signal regulatory protein alpha (SIRPα, also known as CD172a) on myeloid cells. By blocking the SIRPα-CD47 interaction, which cancer cells exploit to evade immune surveillance, DS-1103a acts as an immune checkpoint inhibitor. This blockade enhances macrophage-mediated phagocytosis of tumor cells and is being developed primarily for use in combination with other anticancer therapies, such as antibody-drug conjugates like datopotamab deruxtecan (Dato-DXd) and trastuzumab deruxtecan (T-DXd). The drug is under development for the treatment of advanced solid tumors and is currently in phase I clinical trials. Daiichi Sankyo is the originator and developer of DS-1103a, with AstraZeneca collaborating on its development[1][2][3][4][5][8].
