DS-2741a is a humanized monoclonal antibody that targets and inhibits the calcium release-activated calcium channel protein 1 (ORAI1), which forms the pore of the CRAC (calcium release-activated calcium) channel. ORAI1 is essential for store-operated calcium entry in lymphocytes and plays a critical role in T cell and mast cell activation. By inhibiting ORAI1, DS-2741a suppresses T-cell activation and mast cell degranulation, leading to reduced skin inflammation in animal models of atopic dermatitis. The drug was developed by Daiichi Sankyo primarily for immune-mediated diseases such as atopic dermatitis but development has been discontinued after phase 1 trials[1][2][3][4].
