DS-3078a is an investigational small molecule drug that acts as a dual inhibitor of the mammalian target of rapamycin complexes 1 and 2 (mTORC1 and mTORC2). By binding to and inhibiting both TORC1 and TORC2, it disrupts the PI3K/Akt/mTOR signaling pathway, which is frequently dysregulated in human cancers. This inhibition can result in tumor cell apoptosis and decreased tumor cell proliferation. The drug was developed by Daiichi Sankyo for use in advanced solid tumors and lymphomas. Clinical development included a Phase 1 open-label study to assess safety, tolerability, pharmacokinetics, and preliminary efficacy in patients with advanced cancers who had no further standard treatment options[1][2][3][4][5][10].
