Overview

DS-3939 is a specifically engineered, potential first-in-class antibody-drug conjugate (ADC) targeting tumor-associated mucin 1 (TA-MUC1), a cancer-specific transmembrane glycoprotein overexpressed in many epithelial cancers. The drug consists of a humanized anti-TA-MUC1 antibody (gatipotuzumab) linked via a peptide-based cleavable linker to a potent topoisomerase I inhibitor payload (DXd, an exatecan derivative). Developed using Daiichi Sankyo’s proprietary DXd ADC technology, DS-3939 is designed to selectively deliver the cytotoxic payload to TA-MUC1-expressing tumor cells. It is currently being evaluated in phase 1/2 clinical trials for patients with advanced solid tumors—including non-small cell lung, breast, urothelial, ovarian, biliary tract, and pancreatic cancers—who are not amenable to standard treatments[1][3][4][5][6].
