DS-7080a is a humanized monoclonal antibody developed by Daiichi Sankyo for the treatment of neovascular (wet) age-related macular degeneration (nAMD) and diabetic macular edema (DME). It acts as an angiogenesis inhibitor by selectively targeting Roundabout 4 (ROBO4), a vascular-specific protein involved in stabilizing vasculature during pathological ocular angiogenesis. Unlike anti-VEGF agents, DS-7080a suppresses migration of endothelial cells induced not only by VEGF but also by other growth factors such as HGF and bFGF. Preclinical studies demonstrated that intravitreal administration of DS-7080a reduced choroidal neovascularization and vascular leakage in animal models. Clinical development included phase 1 trials evaluating its safety and preliminary efficacy both as monotherapy and in combination with ranibizumab for nAMD and DME[1][2][3][5].
