DS-8273a is an agonistic monoclonal antibody that targets human death receptor 5 (DR5), also known as tumor necrosis factor-related apoptosis-inducing ligand receptor 2 (TRAIL-R2). By binding to DR5, DS-8273a stimulates apoptosis in cancer cells and has demonstrated cytotoxic activity against a range of human tumor cell lines. It also induces apoptosis in myeloid-derived suppressor cells (MDSCs), potentially enhancing anti-tumor immune responses. The drug was developed by Daiichi Sankyo and has been evaluated primarily for the treatment of advanced solid tumors, including melanoma, colorectal cancer, lymphoma, and other malignant solid neoplasms. Clinical development reached phase 1 trials, where it showed good tolerability but limited objective responses as a single agent[1][2][4][7].
